Myo-inositol is a vitamin-like compound that helps your cells respond to two key hormones: insulin, and FSH, the hormone that ripens eggs in the ovaries. The body makes some from glucose and gets the rest from food. It was once called vitamin B8, though it is not a true vitamin because the body can make it.

It has become one of the most talked-about supplements for PCOS, and doctors in India now recommend it often. This guide explains how it works, what studies show, the famous 40:1 ratio, the usual dose and the side effects to know about.

Why myo-inositol matters in PCOS

In many women with PCOS, cells respond poorly to insulin. This is called insulin resistance. The body makes extra insulin to compensate, and high insulin pushes the ovaries to make more androgens, the male-type hormones. That chain shows up as irregular periods, acne, facial hair, hair thinning and weight that settles on the belly.

Inside cells, myo-inositol works as a messenger. When insulin or FSH knocks on the cell's door, inositol helps carry the message inside. Research suggests women with PCOS often handle inositol differently, so the message gets weaker. Supplementing is meant to strengthen that signal.

Myo-inositol uses: what research shows

  • More regular cycles. Several clinical trials and reviews found improved cycle regularity and ovulation in women with PCOS.
  • Better insulin response. Studies report lower fasting insulin and better insulin sensitivity scores.
  • Lower androgens. Some trials saw falls in testosterone, which may help acne and unwanted hair over time.
  • Egg quality. In fertility clinics, myo-inositol has been studied for egg and embryo quality during IVF.
  • Pregnancy. Doctors have studied it for lowering the risk of sugar problems in pregnancy. This is strictly a doctor-led use.

It is not a weight loss pill. Some women lose a little weight as insulin improves, but food and movement still do the heavy lifting.

Myo-inositol and D-chiro inositol: the 40:1 ratio

Inositol comes in two main forms that work as a team. The body converts myo-inositol into D-chiro inositol using an enzyme switched on by insulin.

  • Myo-inositol dominates in the ovaries, where it supports FSH signalling and egg development.
  • D-chiro inositol is stored more in muscle, fat and liver, where it supports insulin's job of storing sugar.

In healthy blood, the two sit at about 40 parts myo to 1 part D-chiro. Many combination products copy this 40:1 ratio, and a 2019 study comparing different ratios found 40:1 worked best for restoring ovulation. Too much D-chiro inositol on its own may work against egg quality, which is why most doctors prefer myo-inositol alone or the 40:1 blend.

Myo-inositol dose: how much and how long

Question What studies and doctors usually use
Daily amount 2 g twice a day, so 4 g in total, often with 200 mcg folic acid
Form 2 g powder sachets, or several smaller tablets through the day
Timing Split between morning and evening, with or without food
Time to see change At least 3 months. Cycle changes often appear in 2 to 3 months
Egg quality About 3 months, since an egg takes roughly 90 days to mature

Your doctor may suggest a different amount, especially if you are trying to conceive. Always follow their advice over any general guide.

Myo-inositol side effects

Myo-inositol is one of the gentler supplements. At the usual 4 g a day, side effects are uncommon. At much higher doses, around 12 g a day, people report:

  • Nausea, gas or loose stools
  • Tiredness, headache or dizziness

If you take medicine for diabetes, mood or fertility, or if you are pregnant, check with your doctor before starting. A common question is whether myo-inositol causes weight gain. There is no good evidence that it does.

Myo-inositol vs metformin

Both target insulin resistance, which is why they are often compared. Some trials found similar improvements in cycles and insulin markers, with fewer stomach side effects for inositol. Metformin, though, is a prescription medicine with a much larger body of evidence for blood sugar. Many doctors use both together. Never swap one for the other without your doctor.

Myo-inositol rich foods

  • Oranges, mosambi and other citrus fruits
  • Kharbooja (cantaloupe)
  • Beans, rajma and chana
  • Whole grains such as oats and wheat bran
  • Nuts, especially almonds and walnuts

A normal diet gives less than 1 g a day, well below study amounts. Food is the foundation, but it cannot reach supplement doses on its own.

Where Purezen HerBalance fits in

Purezen HerBalance is a 20-ingredient capsule that combines Myo-Inositol and D-Chiro Inositol with Chasteberry, Shatavari, Ashoka, NAC, Cinnamon, L-Methylfolate, Chromium and Vitamin D3. These are herbs and nutrients traditionally used to support hormonal balance, a more regular cycle and healthy metabolism in women.

HerBalance is a broad daily formula rather than a single high dose of inositol. If your doctor has prescribed a specific inositol dose, keep to it and ask before adding anything. Purezen is FSSAI registered and HerBalance uses a vegetable capsule shell. It works alongside food, movement and your gynaecologist's care, not in place of them. NAC, one of its ingredients, is covered in our NAC guide.
